it is pretty easy to do, just did mine, I've resprayed the entire car and had to remove those moldings and stick em back. you only need to put some adhesive sillicone on both ends, just a dab big as a pea. Then you put some adhesive tape over both ends until it cures.

Does the water leak inside the cabin if you remove the weatherstripping?
nope. Except if the seams around your back glass of windshield are cracked (wich is nearly impossible with factory installed glasses) Basicly this molding is there to hide the welding spots for the roof metal sheet.
