This article http://cjsupra.kendra.com/mkv.html suggests that the next Toyota Supra will use the existing Iron block inline 6 (making 330hp). So if Toyota is sticking with an iron block for the next Supra then I doubt that Lexus will switch to an Aluminum block anytime soon..

This is an outdated post, the new Supra, likely to be renamed the 4000GT, will have an all aluminum V8. I don't know if it will be a TT version of the V8, but it will definitely have a V8.
You can check out Supra forums for more recent updates. My guess is that it'll show up at the NAIAS next year - at least, I hope so.
I just discovered that the Toyota V6, that they use in the ES300 and RX300 (and other cars), was an aluminum block. I wonder if they can pop that engine in the IS - less weight and more power, with a supercharger.

Will the Toyota 4000GT be Japan's first V8 powered Sports car ever?

Not if the NSX comes out first.. I'm sure the 4000GT will be based on the Lexus V8.. probably destroked for more rev potential..
